OK, it isn't a sign, but last year at the Disney Marathon my wife and two friends and I crossed the start 45 minutes after the start time.
We were the last to get going, due to a detour getting to the start.
At the first mile marker, instead of getting a cheer, one of the volunteers looked at the 4 of use and said
"You are NOT making it"

We passed over 3000 people that day.

Not a funny one, but at IMCDA in 08 passed a sign that said

"Pain never hurts as much as quitting" Helped get me through the next mile at least.

Saw a couple at the Woodlands IM (spectating not competing) that made me laugh...

"DON'T TRUST A FART!" mile 14 of the run
"Concentrate on your bloody nipples to take your mind off your sore legs!"
"Just keep falling forward!"

P.S. Love the "I don't do triathlons I do a triathlete" sign... my ex  had one similar at my first tri!
