Bike with Sora shifters and the rest Tiagra components. If I change the front and rear deraileur to Dura-Ace will it really matter?

I'm not sure whether Shimano components are compatible between levels--maybe somebody else knows.

I'd do the shifters first, that's probably where you'd notice the biggest difference.  Rear derailleur would be next, then front.

assuming tiagr and sora are compatible with the 105, ultegra and DA stuff,

i would go for a full rounded 105 group, or just the shifters right now and maybe rear derailer.

after 105 your not going to notice ANY diff in how it works at all, and frankly will prob last a lot longer than DA stuff will, although it will be a tiny bit lighter.

just my thoughts though.

Sounds like a bit of overkill.  What kind of bike are you talking about?  I suspect there might be other ways to spend your money that would have more of a performance impact on your bike. 

I've got a combination of 105 and Ultegra on my Cannondale and am perfectly happy with it.  I'd put my money towards the purchase of a new bike before I'd upgrade to DA.

Yes, it would. That said, upgrade your bike with one that comes with 105 or Ultegra long before you do this. For one, it'll be cheaper.

Sora and Tiagra are both 9 speed drivetrains.  10 speed 105, Ultegra, and DA wouldn't be compatable.  You could check and see if older 9 speed 6500 Ultegra or 7700 DA components would work (they should), but you'd be better off changing the whole group.  9 speed stuff should be very reasonable, though.

If you have a high end frame and low end components, sure. If you have an entry level frame, not worth it at all.

Sora and Tiagra both work, in fact they can work quite well for a long, long time. As pointed out they are just 9 speed and not 10, but the only thing you get with 10 speed is slightly more granular gear selection. I was still rocking a 9 speed up until two weeks ago and I only upgraded because of a smokin deal on a new bike.

Save your money toward a new bike and just ride a ton. Working on the engine is way more important than Dura Ace this, Red that and a fancy set of Zipps that sets you back a pay check. You'll find it quite satisfying passing a $4k bike on your Tiagra/Sora equipped one.
