I am not 300+ anymore so I can't speak specifically to your question, but...
I bought my Cervelo S5
(aero but not tri bike
) when I weighed about 315, I did my research and found that Cervelo had no weight limit on the frame - so I would say that there are likely options that would be okay from a carbon frame standpoint. They do say that certain components may have weight limits so that would be something to think about.
As for the tribike position, I just got a Cervelo P2. I have not had an in depth fitting done yet as my fitter and I decided, due to my schedule, that we would do the fitting when I brought it back for a "10 hour checkup". At about 250 lbs with a "quick"
(still over an hour
) fitting and about 150 miles I notice...

) There are times that my belly feels like it is somewhat in the way. This makes it somewhat difficult to make a strong climbing effort and I think it leads to a bit of out of breath feeling, but it seems to be diminishing a bit so it might just be function of getting used to the different position.
b.
) My arms, specifically my tricep area, tend to get sore rather quickly. This too seems to be getting better, but the first few rides I was only able to maintain aero for about 5 minutes at a time. I wonder if core strength vs. size may add to this, but that is strictly theorizing. As of last night I was able to ride about 75% of my ride in aero but still had to do some fidgeting around to keep my arms happy.

) I am taking a big hit on climbing. It just feels SO much harder. Not sure if being big has a larger effect on this than normal, but hills are currently a bit discouraging.
Not sure if these help at all and my opinions definitely may change once I have my fitting, but mostly I would recommend talking about your concerns with your LBS and taking a good length test ride before making a decision to buy something.
OH...And most importantly...CONGRATZ on dropping 140 lbs!!! That is awesome!!!
