Longer rides I like my shorts with the most padding but just a couple of hours or so I will use tri shorts or if its just a hour or so I will just wear compression shorts so my tail toughen ups more.

I started riding on fully padded bike shorts.

I realized quickly that the chamois was not very comfortable.

I ride completely in tri shorts now.  Enough padding to keep the friction down, but not so much that you feel like you're wearing a diaper.

Tri shorts for me.  I find that bike shorts with thick padding are not comfortable when riding in the aero position.  The padding bunches up in my crotch.  I also use Chamois Butter in the tri shorts for long rides.  I just completed IMFL in tri shorts. 

Comfort on the bike is also a function of bike fit and time in the saddle.  Ride lots and you'll be lots more comfortable.

I use regular cycling shorts for all my rides, and they've always provided more than enough comfort.  The more I ride, though, the less I notice whether the padding is or isn't enough - I suppose it's because I'm accustommed to being in the saddle so much that it doesn't matter to me anymore.
